Experimental method for efficiently breaking muscle tissue samples using TL2010S tissue disrupter

Through a large number of sample crushing tests, Ding Yiyuan Technology found that animal tissue samples are more difficult to break than other samples, especially some connective tissue, heart muscle tissue, and lung tissue. Because of the high toughness, it is generally difficult to use manual or conventional samples. The tissue homogenizer is completely broken. Because no matter whether it is hand-grinding or tissue homogenizer, not only the treatment flux is small, easy to be contaminated and difficult to clean, the sample after treatment is not uniform and detailed, and the requirements of subsequent experiments are not met. In the test, Dingyiyuan Technology used TL2010S tissue crusher ( also known as TL2010S flux tissue grinder ) to break muscle tissue samples, and the crushing effect was uniform and detailed, which met the requirements of subsequent test, component analysis and nucleic acid analysis. The operational points are as follows:

1. Put the muscle tissue coarse sample into 5ml ordinary round bottom or special grinding tube, and the sample volume is about 1/3 volume of the single tube;

2. Insert 3 stainless steel grinding beads with a diameter of 8mm in each centrifuge tube, cover the tube cover, and insert the centrifuge tube into the adapter symmetrically and fix it to the two rocker arms;

3. Set the parameters of the TL2010S tissue crusher to 1800 rpm , cycle 5 times, stop grinding 10S per grinding for 60 seconds (ie intermittent grinding for 5 minutes);

Use the TL2010S tissue crusher to crush different volumes and quantities of samples at one time. Not only is the operation flexible and convenient, but also avoids cross-contamination between samples, eliminating the tedious task of cleaning the instrument.


In the test, the choice of the centrifuge tube also has a certain effect on the effect of sample breakage. We know that the TL2010S tissue breaker can also be milled with multiple 1.5/2.0ml centrifuge tubes, but this type of centrifuge tube has a smaller capacity than the coarse sample that is crushed in the experiment, and it can accommodate the grinding beads. Small, the impact force is also very limited, so it is only suitable for batch processing of a large number of small samples. With a 5ml centrifuge tube, the amount of sample placed in a single tube is not only increased, but larger diameter beads can be placed to create a greater impact force for better grinding results.


Another key factor is intermittent grinding, which is one of the special features of the TL2010S automatic tissue crusher , which also has a certain improvement effect on the crushing effect. Because the intermittent time is set, the sample can be effectively reduced in the continuous high-speed grinding and breaking due to rapid temperature rise, which provides guarantee for the smooth progress of subsequent experiments. If it is combined with liquid nitrogen for low temperature treatment, it can reduce the temperature rise of the sample more effectively, which is good for temperature-sensitive experiments such as RNA extraction.
